If we're talking sitcoms, I have to give the nod to The Honeymooners as the GOAT. They established the genre for everybody else.

For television I like The Dick Van Dyke Show. It had the perfect premise-the office and home life of a comedy writer. And the cast and production team turned out to be legendary.
Some Like It Hot is the AFI best comedy film. I also love Operation Petticoat and Ruthless People.

I'm glad you mentioned those two.
Dick Van Dyke's physical comedy was unmatched during that era.
Barney Miller had such a great range of characters.
Strictly in terms of humor the first two that stand out to me are The League on t.v. and Something about Marry in theater

Loved his show when I was a kid. His was the first celebrity death that really saddened me even though I was only 10.
